驼源肠道外致病性大肠杆菌分离鉴定及生物学特性分析Isolation,identification and analysis of biological characteristics of pathogenic Escherichia coli isolated from camel

摘要(Abstract):
从内蒙古死亡双峰驼病料中分离致病菌,对其进行生物学特性分析,为治疗和预防该类疾病提供依据。采集死亡双峰驼的肺脏、肝脏、肾脏、心脏和肠管等组织器官,进行病原微生物的分离,观察病原菌的培养特性和染色特性,分析病原菌生化特性。通过与5种肠道内致病性大肠杆菌的核酸比对进行分型鉴定,对该病原菌进行基因组测序,并对其致病性、药物敏感性和有关毒力基因进行分析。结果显示,从双峰驼病料中分离出1株革兰阴性杆菌。该菌在LB琼脂培养基中形成白色、椭圆、光滑的菌落,在伊红美蓝培养基中菌落呈现亮金属色光泽,在哥伦比亚血平板上有明显的溶血环。经全自动生化鉴定仪鉴定,该菌属于大肠杆菌属。PCR结果显示该菌株不属于5种常见的肠道内致病性大肠杆菌,测序分析确定该病原菌为肠道外致病性大肠杆菌。对其进行动物回归试验发现,该菌株有强致病性。药敏试验结果显示,该菌对β-内酰胺类、四环素类、氨基糖苷类、磺胺类、喹诺酮类、多肽类、氯霉素类药物呈现多重耐药,其携带的耐药基因有TEM、tet(B)、aph(3)-1、aac(3)-1、sul-1、sul-2、qnrB、cmlA、cat-1。毒力基因分析显示其携带的毒力基因有irp2、fyuA、iucA、FimA、FimC、iss、colBM、sheA。结果表明,引起双峰驼死亡的病原菌为肠道外致病性大肠杆菌,该菌株携带多种耐药基因和毒力基因,表现出多重耐药和强致病性特征。
