犬颈总动脉粥样硬化致狭窄模型的建立Establishment of canine model of common carotid atherosclerosis induced stenosis

摘要(Abstract):
通过使用球囊损伤联合高脂饲料喂养的方法建立犬颈总动脉粥样硬化致狭窄模型,并探讨介入治疗机制。选取1周岁雌性比格犬6只,分为对照组(Ⅰ组)和试验组(Ⅱ组),每组3只,试验组给予高胆固醇饮食,第4周时对犬颈总动脉实行球囊损伤术,记录试验过程中其体质量、血管内径及血脂水平(TC、TG、HDL-C和LDL-C)变化情况,观察颈动脉管腔病理组织切片、脂肪浸润及弹力纤维变化等情况,对照组进行假手术并给予基础饮食。结果表明,试验组体质量随测量时间呈现增长趋势,与对照组差异极显著(P<0.01);球囊损伤术后犬存活率100%,血管内径极显著性降低(P<0.01),且呈中度狭窄;与对照组相比血清水平HDL-C含量变化不显著,TC、TG和LDL-C含量显著升高(P<0.05);20周与第1周(基础值)相比TC含量上升2.0倍,TG含量上升1.6倍,LDL-C上升2.5倍;组织切片可见血管管腔变狭窄,内膜至中膜有脂质沉积,大量泡沫细胞形成,弹力纤维断裂消失。本试验通过饲喂高脂饮食联合球囊损伤可成功使犬颈总动脉形成动脉粥样硬化并导致狭窄,该模型具有成活率高、建模时间短、生理和病理状态相似的特点,可用于研究犬颈动脉粥样硬化导致动脉狭窄的治疗方法。
