维甲酸对成肌细胞细胞周期进程及凋亡的影响Effects of all-trans retinoic acid on cell cycle progression and apoptosis of C2C12 mouse myoblasts

摘要(Abstract):
成肌细胞的周期进程及凋亡对骨骼肌的损伤修复至关重要。本研究旨在细胞水平探讨全反式维甲酸(ATRA)对C2C12细胞周期进程及凋亡的影响。试验组C2C12细胞用100 nmol/L ATRA处理48 h,对照组不予处理。用MTS、Ki67及EdU法检测ATRA对C2C12细胞增殖的影响,通过Western blot检测ATRA对C2C12细胞中增殖相关蛋白CDK2、CDK4、CDK6以及细胞周期蛋白D1 (Ccnd1)、Ccnd2、Ccnd3和Ccne1表达的影响,并检测ATRA对Bcl-2、Bax、Caspase-3等凋亡相关蛋白表达的影响,最后通过流式细胞术检测细胞周期进程以及凋亡率。结果显示,与对照组相比,100 nmol/L ATRA可促进C2C12细胞增殖,并且上调上述几种增殖相关蛋白的表达水平。ATRA促进C2C12细胞从G1期进入S期。此外,ATRA处理使C2C12细胞Bcl-2显著升高,而Bax及活化的Caspase-3显著降低,从而显著降低了细胞凋亡率。综上所述,100 nmol/L ATRA处理48 h会促进C2C12细胞的增殖并抑制细胞凋亡,对骨骼肌的损伤修复具有潜在作用。
